The melanocortin peptides are studied through a family of five receptors, and the practical difference between the compounds in this category is which of those receptors each one engages and how selectively.
Melanotan II is a cyclic alpha-MSH analogue and the broader-acting of the two - it is the earlier compound, and the one most of the melanocortin literature is built on. PT-141, also called Bremelanotide, was engineered from that work to act more selectively through central receptors. KPV, listed separately under inflammatory research, is a fragment of the same parent molecule but is not reported to produce the pigmentation effects associated with the fuller analogues.
